The influences of two dyes, methylene blue and acridine orange, were examined on the contraction in the vas deferens from guinea pig. Methylene blue in low concentration increasedphasic contraction but decreased tonic contraction developed by K. It also potentiated the contraction by ACh. While, its high concentrations reduced phasic contractions by K and ACh. The effects of acridine orange were similar to those by methylene blue. High concentration of acridine orange showed an irreversible inhibition against the contraction. These results suggested that dyes in low concentration inhibited an energy-dependent action and that those in high concentration besides showed the inhibition with energy-independent one. Furthermore, high concentration of methylene blue itself produced a contraction, and it seemed to depend on Ca release.
