It has been accepted that SHR is a useful animal model for the study of human essential hypertension, so many investigations have been made to clarify the hypertensive mechanism of SHR. Thereby, it is reported that a cause of hypertension of SHR may be due to the increased response of blood vessel to vasocontractile agents. Whether this increased response is due to the hereditary or the secondary from high blood pressure has not, however, been fully clarified. In the present study, the author studied the responses of aortas and portal veins from stroke-prone SHR (SHRSP) to the vasocontractile agents from the standpoint of aging and compared the results with those of Wistar Kyoto rats (WKY). Aorta : The contractions by noradrenaline (Nad) and phenylephrine (PhE) increased with age more remarkably in WKY than in SHRSP, being larger in SHRSP than in WKY at 1.5 and 3 month (M). But those were not different between both at 6 and 12 M. The contractions by K^+, Ca^<2+>, serotonin, prostaglandin F_<2α> increased with age more remarkably in SHRSP than in WKY. The contractions by those agonists were larger in SHRSP than in WKY at any age tested. Although the contraction by angiotensin II (Ang) was larger in SHRSP than in WKY at 1.5 M, those at 3,6 and 12 M were larger in WKY than in SHRSP. The pD_2 values for Nad, PhE and serotonin were not different between the both at any age tested. The pD_2 value for Ang in SHRSP was not different from that of WKY at 1.5 M, but those in SHRSP were smaller than those in WKY at the other ages tested. Portal vein : The contractions by Nad, PhE, Ca^<2+>, K^+, Ang, serotonin, acetylcholine and prostaglandin F_<2α> increased with age, more significantly in SHRSP than in WKY. Thereto, the contractions by those agonists were larger in SHRSP than in WKY at any age tested. The pD_2 values for Nad, PhE, Ang and acetylcholine were not different between the both at any age tested. These results suggest that the increased response of blood vessels from SHRSP to the vasocontractile agents might be due to the hereditary in large part and in small part due to the secondary from the high blood pressure.
