An application of the x, y and Y' chromaticity coordinate system to the quantitative histochemical measurements was presented on the bovine nuchal ligaments. In order to make a standard for comparison of elastic fibers at various sites, spectral transmittance measurements were carried out on the Weigert-stained elastic fibers, and the values were processed to give a series of the chromaticity coordinates. The locus of the coordinate was spiral around the C (white light) axis in the color space. The brightness was linear to the minimum transmittance values within the experimental error. Whenever one wants to compare an amount of the Weigert-stained elastin to the other, this locus curve will be useful not only for calibration line but also for identification of materials. Limitations and conditions for microscopic spectrophotometry with the chromaticity coordinate system were discussed.
