Three rat strains, M-SHRSP, SHRSP, and SHR B_2 displaying extremely severe, severe, and moderate spontaneous hypertension, respectively, and normotensive WKY were used to determine plasma aldosterone concentrations (PAC), plasma renin concentrations (PRC), and the morphology of the zona glomerulosa of the adrenal gland. The results were as follows : 1. PAC increased significantly in M-SHRSP over that in WKY from 10 weeks of age in males and from 12 weeks in females, i.e. at the same time blood pressures went above 230 mmHg. In the males at 12 and in the females at 15 weeks PAC values were about six times those of similarly aged WKY. In SHRSP the PAC increased more slowly and reached only 2.5 times the WKY values at 25 weeks. SHR B_2 showed no significant increase in PAC. 2. Similarly to the increases in PAC, significant increases in PRC for male M-SHRSP took place at 10 weeks and for the female at 12 weeks. SHRSP PRC increases occurred much earlier than did the PAC increases, being seen at 12 weeks in males and at 20 weeks in females. SHR B_2 PRC values remained practically unchanged. 3. There were positive correlations between blood pressure and PAC, between blood pressure and PRC, and between PRC and PAC in mature M-SHRSP with blood pressures above 210 mmHg. 4. a) Adrenal weight ratios (to body weight) for M-SHRSP were significantly higher than for WKY after 11 weeks in males and after 15 weeks in females ; this was also true for male SHRSP after 15 weeks. b) The zona glomerulosa was significantly thicker in M-SHRSP than in WKY after 10 weeks in males and after 15 weeks in females ; the same for SHRSP being after 15 weeks in males and after 25 weeks in females, respectively. The cells in the thickened zona glomerulosa stored lipids (mainly cholesterine ester) and appeared clearer and enlarged. 5. The increases in PRC, the obvious thickening of the zona glomerulosa, and the increases in PAC gradually developed after blood pressures went above 210 mmHg. 6. Based upon previous reports on M-SHRSP characteristics plus the findings in this study it may be concluded that M-SHRSP shows symptoms and progress in hypertension that are very similar to those found in human juvenile malignant hypertension.
