A principle to correct scattered light effect on the microscopic spectrophotometry of translucent preparations is described. In the Weigert's resorcin-fuchsin staining, elastic fibers showed a hue from blue to reddish purple. Calculation of elastin concentration in vascular media in relation to SHR survey revealed pathological changes of elastic fibers quantitatively as well as qualitatively. The approximation method indicated that the elimination of scattered light produced mainly increases in the lightness, Y' and W. This study also gave a basis for the comparison of the chromaticity coordinates at the Y' = 100% plane. An increase in the color distance caused by the correction did not exceed the ones among specimen differences.
