Changes in cerebrospinal fluid pressure (CSFP) and blood pressure (BP) were measured in 10 adult Wistar-Kyoto rats (WKY) and 20 adult stroke-prone SHR (SHRSP). The CSFP measurement was made at the cerebello-medullary cistern by using a thin needle inserted through the occipito-nuchal fascia. The needle was connected to a pressure transducer via a thin polyethylene tube. Major difficulties in measuring CSFP in SHRSP include cerebral damage and brain extrusions resulting from their abnormally high CSFP. The CSFP of adult SHRSP with BP of ca 240 mmHg is significantly higher than that of adult WKY with normal BP of around 130 mmHg. BP and CSFP changes resulting from hydralazine hydrochloride administration (10 mg/kg body weight) were qualitatively similar. The general pattern of CSFP was shown as a slight lowering at first, then a peak representing an instantaneous increase which lasted for several minutes, and finally a falling phase which continued until death. These CSFP changes are discussed in relation to a compensatory function related to BP. A two-step feedback schema is also proposed as a model explaining the CSFP regulatory mechanism.
