Using 10-week-old male M-SHRSP, a strain of rat with severe precocious hypertension, zona glomerulosa hypertrophy and increases in plasma aldosterone concentrations (PAC) were investigated following administration of metopirone, spironolactone, captopril, or guanethidine sulfate. Results were : (1) After administering metopirone, spironolactone or captopril for 2 weeks, PAC values decreased significantly while the administration of guanethidine prevented an increase in PAC values. (2) Blood pressures decreased significantly only in the group given captopril. (3) Captopril reduced plasma renin concentrations but metopirone increased these values. (4) The adrenal weight to total body weight ratio decreased in the group given captopril, but increased significantly in the groups administered metopirone or guanethidine. (5) Adrenal zona glomerulosa hypertrophy (thickening) was seen in the group administered metopirone, but for the other groups there was a tendency for the adrenal zona glomerulosa to be thinner than those of non-treated controls. This thinning was particularly significant in the captopril group, with values nearly equalling those of normotensive Wistar-Kyoto rats. (6) Administering captopril or spironolactone for two weeks remarkably prolonged the life spans. Overall, these results suggest that deterioration of the renin-angiotensin system increases hypertension in M-SHRSP, but that aldosterone, itself, is not an important factor.
