Strongly calcified aorta found in a SHR specimen was examined with four different methods for calcium detection in connection with the normal rat group. Four histochemical methods used were as follows ; fluorescence method with 8-hydroxyquinoline, a modification of Okamoto's quinalizarin method, GBHA method and lead transposition method. The sensitivity was highest in the GBHA method, and the fluorescence method showed the least sensitivity among them. For quantitative estimation of calcium, the quinalizarin method and the lead transposition method were satisfactory because of their stabilities of the color. The estimated calcium concentrations in the normal group were 7-18 mM in the intima and the inner one-third of the media. Strong calcification in a SHR specimen was 1.5×10^2 mM.
