The effects of hypotonicity on bullfrog sartorius muscle cells were examined both structurally and by electrophysiological methods. As the bullfrog sartorius is convenient for physiological studies, some of the mechanical and structural characteristics were also investigated. The bullfrog sartorius contained nearly 1500-1600 muscle cell fibers, 12% of which were thicker (cross section area : 12-10.7×10^<-10>m^2) and the rest, thinner (10-7×10^<-10>m^2) muscle cells. With the use of chloride-sensitive microelectrodes, the calculated internal Cl^- concentration shifted from 0.004mM at the beginning of the experiment, and to 0.038mM after 1 hr soaking in distilled water. The membrane potential of the hypotonically swollen muscle cells showed hyperpolarization below the resting level. This apparent membrane potential shift corresponding to the hypotonic environment is estimated as the increase of the chloride conductance caused by the structure deformation of muscle cell membrane.
