ハマウツボ科のニクジュョウ(Cistanche salsa(C.A. Meyer) G. Beck)は,日本では強壮作用を有する和漢薬として使用されている寄生植物である。我々は前回の研究において,ニクジュョウ抽出物がヒトB細胞株のBALL-1細胞に対して,わずかな細胞増殖活性と共にIgM産生増強作用を有することを明らかにした。本研究では,この抽出物がヒトバーキットリンパ腫由来の細胞株であるNamalwaには逆の作用を有することを示す。分子量3,500Daカットオフの透析膜を使用して得られたニクジュョウ抽出液の透析物(CSD)は,Namalwaに対して細胞増殖抑制作用を示した。そしてこの増殖抑制作用をDNAの断片化とサブG_1細胞の割合により評価した結果,CSDで処理された細胞はアポトーシスの特徴を示し,核内DNAはラダー状に断片化し,サブG_1細胞の割合も高くなっていた。これらの結果より,Namalwaに対するCSDの作用は,B細胞の活性化に関与しているものの,B細胞の分化に必要なサイトカインや細胞間相互作用のような他のシグナルの欠損によりアポトーシスが起こることが示唆された。
