From the results of earliear papers in this series on cell bound isoamylase formation by Aerobacter aerogenes, it was considered necessary to satisfy the following conditions for cultivation. That is, in order to obtain higher isoamylase activity per unit cell weight, a cell concentration of more than 10g/l in the fermentated broth was needed. The presence of an excess carbon source (soluble starch) in broth having over 5-6g/l of cell concentration caused an oxygen deficiency. Isoamylase formation in low dissolved oxygen concentration decreased in comparison with that in high DO.For these reasons, continuous cultivation with control of the dissolved oxygen concentration in the broth was used for cell-bound isoamylase formation of this strain by regulating the rate of aeration and feed.The control of the dissolved oxygen by the above means was successful. The cell growth and cell bound isoamylase formation were stable during continuous cultivation and the carbon source in the feed medium was completely consumed by cells. With respect to cell bound isoamylase activity, the continous cultivation gave about the same value as in the feeding cultivation.
