Morphological Differenctitaion in Relation to Cephalosporin C Synthesis by Cephalosporium acremonium

To ascertain a relationship between cephalosporin C synthesis and morphogenesis, the morphological variaton of Cephalosporium acremonium was investigated in detail through all satages of fermentation. Morphological differentiation of fliamentouws hyphae into highly swollen hyphal fragments preceded teh onset of cephalosporin C synthesis. Depletion of glucose in the medium caused hyphal fragmentation and active production of cephalosporin C. The antibiotic was produced along with marked fragmentaion of highly swollen hyphal fragments into spherical unicellular arthrospores.A one-to-one correspondence was observed between the final percentage of spherical uniclellular arthrospores among various morphological cell types and the final titer of cephalosporin C when various concentrations of DL-methionine, D-methionine and L-methionine were added to the cultuer medium initially. The stimulative concentration of methionine for spherical unicellular arthrospore formaiton and the antibiotic of methionine for spherical unicellular arthrospore formation and the antibiotic produciton was lower in D-methionine than in L-methionine.A highly productive strain, CW19,possessed a greater capacity than the parent strain M8650 for forming highly swollen hyphal fragments, although no significant difference was observed in the final amount of the spherical unicellular arthrosopres.
