Simulation of Fed-batch Culture for Glutamic Acid Production with Ethanol Feeding by Use of Regression Analysis

A fed-batch culture of Brevibacterium divaricatum NRRL 2311 producing glutamic acid with ethanol feeding was investigated to determine the time course of feed rate suitable for glutamic acid production. The results of fed-batch cultures at constant concentrations of ethanol in the range of 1 g/l to 50 g/l revealed that cell growth was suppressed and the early production of glutamic acid was promoted by high concentration of ethanol. The duration of glutamic acid production was prolonged was by maintaining a high ethanol concentration of 25 g/l or 35 g/l. When ethanol concentration was increased suddenly from 5 g/l to 25 g/l during fed-batch culture, glutamic acid production reached 26 g/l, much higher than when ethanol concentration was kept constant. To predict numerically the time course of the fed-batch culture, a simulation algorithm involving regression analysis was developed.
