Effect of Glucose and Methionine Consumption Rates on Cephalosporin C Production by Cephalosporium acremonium

Kinetics of cephalosporin C production by Cephalosporium acremonium M8650 was investigated in a chemically defined medium. The roles of sugars, glucose and sucrose, and _DL-methionine, the components of the production medium, were investigated and the relationship between the consumptioin rate of substrates and the production rate of cephalosporin C was quantitatively studied. Lumped sucrose supply could be replaced by intermittent glucose supply at the appropriate rate for the produciton of cephalosporin C, whereas extra addition of _DL-methionine in the production phase did not increase the accumulation of the antibiotic. In a glucose-limited continuous culture, the production rate of cephalosporin C was closely dependent on both the rate of microbial growth and the utilization rate of glucose but not on that of _DL-methionine. Furthermore, experiments indicated that the maximum production rate would be obtained with an extremely low dilution rate. Fed-batch cultures with exponential feed of glucose, which were conducted to determine the production rates at very low growth rate, showed that maximum productivity occurs when microbial growth does not apparently occur.
