Continuous Production of 12-Ketochenodeoxycholic Acid in a Column Reactor Containing Immobilized Living Cells of Brevibacterium fuscum

To prevent the over-reduction of the product to cholic acid in the production of 12-ketochenodeoxycholic acid from dehydrocholic acid by Brevibacterium fuscum DC33,we studied the optimal conditions for the continuous production in a column reactor containing living cells of the microorganisms immobilized with Κ-carrageenan. The highest yield, 80%, was obtained at pH 7.0,22℃ and gel-basis time factor of 2.1 g wet gel・hr/ml using 0.5 g/l of the substrate. The column was stable for more than 370 hr. The accumulation of cholic acid was 0.05 g/l, and that of the intermediate, 7,12-diketo-lithocholic acid, was almost negligible.
