Modeling of Cephalosporin C Production and Its Application to Fed-batch Culture

A kinetic model for the production of cephalosporin C by Cephalosporium acremonium was proposed. In developing the model, morphological differentiation, induction of cephalosporin C production by endogenous methionine, and catabolite repression by glucose were considered. Parameters in the model were determined by a modified simplex method so as to minimize th differences between the experimental and calculated values. The model and its parameters were assessed by simulating various characteristic batch cultures. Then it was applied to fed-batch culture to find the best mode of feeding for improvement of antibiotic production. This predicted mode was tested in fed-batch culture with feeding of glucose and methionine and resulted in a substantial increase in antibiotic production of about 30% over that of batch culture.
