Growth of Curvularia lunata Spores into Mycelial Form within Various Gels, and Steroid 11β-Hydrooxylation by the Entrapped Mycelia

Spores of Curvularia lunata were entrapped aseptically with photo-crosslinkable resin prepolymers, water-miscible urethane prepolymers or several kinds of polysaccharides. The entrapped spores were grown in situ for 60 hr in potato-dextrose broth containing Reichstein's Substance S as an inducer of the steroid 11β-hydroxylation system. The entrapped mycelia thus obtained hydroxylated Reichstein's Substance S to from hydrocortisone in a buffer containing 2.5% of dimethylsulfoxide. Of various entrapment methods tested, entrapment with a hydrophilic photo-crosslinkable resin prepolymer, ENT-4000,gave the highest activity, which was comparable to that of the free mycelia. The stability of the enzyme system inside the mycelia over repeated reactions was improved significantly by entrapment.
