Production of D-Amino Acid Oxidase by Candida tropicalis : Studies on the Utilization of Hydrocarbons by Microorganisms (XVIII)

Production of D-amino acid oxidase by petrochemical-utilizable yeasts was examined. Of five strains of n-alkane-utilizable yeasts and three strains of methanol-utilizable yeasts tested, Candida tropicalis was found to be most suitable for D-amino acid oxidase production. Glucose and n-alkane mixture (C_<10>-C_<13>) were good carbon sources for the enzyme production by this yeast. Addition of DL-amino acids, such as DL-alanine, to the medium significantly enhanced the enzyme formation. After the optimization of culture conditions, the maximum activity obtained was about 88 units/mg dry cell in glucose-groun C. tropicalis cells, indicating that the yeast would be a good source for D-amino acid oxidase production. Crude D-amino acid oxidase form glucose-grown C. tropicalis cells exhibited the following properties : Apparent K_m value for D-alanine, ca. 14mM; optimal pH, 9.0; optimal temperature, 37℃. A large part of the D-amino acid oxidase produced in glucose-grown cells of C. tropicalis was located in a microbody fraction, as in the case of n-alkane-grown cells.
