Immobilization of Invertase by the Use of Photo-crosslinkable Resin Oligomers and Properties of the Immobilized Enzyme

This paper describes a novel convenient method for entrapping enzymes or microbial cells by the use of photo-crosslinkable resin oligomers, such as polyethylene glycol dimethacrylate. Invertase, used as test enzyme, was entrapped successfully and easily in the resin film formed by a few minutes' illumination with a chemical lamp (λ_<max>, 360 nm). The activity of the enzyme thus entrapped was 40 to 50% of that of the native enzyme. The relative activity of the enzyme entrapped in the film was affected by the proportion of water, enzyme and oligomers in the immobilization system, the ratio of oligomer containing one functional group to that containing two functional groups, and chain length of the oligomers, but not by the illumination time. The entrapped in vertase showed almost the same V_<max> value and about 5 times higher K_m value compared with the native invertase. The pH and temperature dependencies of the enzyme reaction were also compared between the entrapped and free enzymes. The immobilized enzyme was stable for at least 30 batch reactions (total operation period, 5 hr).
