For the purpose of the synthesis of atisirene (1) and isoatisirene (2), intramolecular Diels-Alder reaction and double Michael reaction were studied and the stereocontrolled construction of the spiro fused bicyclo[2.2.2]octane ring system was achieved by the latter novel method. (1) Intramolecular Diels-Alder Approach Intramolecular cyclo-addition of the triene (7), derived from a tetrahydroisoquinoline derivative (4) gave the tetracyclic compounds (8A and 8B). However the reactions of the properly substituted trienes (9, 11, 12 and 13) were unsuccessful. (2) Intramolecular Double Michael Approach The kinetic enolates from the cyclohexenones (19 and 32) possessing the α,β-unsaturated ester moiety caused an intramolecular double Michael reaction producing stereoselectively the spiro fused bicyclo-[2.2.2]octanes (17 and 33), respectively. Therefore the suitably substituted unsaturated ester (42) was prepared stereoselectively from the dienone (36) via the 1,6-conjugate addition, hydrocyanation and Birch reduction. The double Michael reaction of (42) assembled the tetracyclic compound (44) as a single stereoisomer. Conversion of (44) into the natural product is in progress.
