Preparation and Validation of an Antiserum Specific for Non-Derivatized Gibberellins

• An antiserum recognizing free gibberellins (GAs) was prepared by immunizing rabbits with a GA_4-BSA conjugate. A radioimmunoassay (RIA) and an enzyme-linked immunosorbent assay (ELISA) were set up using this antiserum. This antiserum showed high cross-reactivity to the so-called active GAs, such as GA_1, GA_3, GA_4 and GA_7. The range for measurements of these gibberellin extended from 30 fmol to 3 pmol in both RIA and ELISA. Extracts from immature seeds of P. vulgaris were subjected to determination of GA_1 by RIA and GC/SIM. The two assays provided similar results, indicating the high degree of reliability of the immunoassay.
