Inhibition of Cyclic AMP Phosphodiesterase by Lignans and Coumarins of Olea and Fraxinus Barks

Studies were conducted on adenosine 3',5'-cyclic monophosphate (cyclic AMP) phosphodiesterase-inhibition by phenolic constituents derived from Olea and Fraxinus barks (Oleaceae), i.e., 38 derivatives of lignans having 2,6-diarylated 3,7-dioxabicyclo[3.3.0]octane ring and 17 derivatives of di- and tri-hydroxylated coumarins. High inhibitory effect was shown on cyclic AMP phosphodiesterase by lignan components of bark of Olea europaea L. and O. africana MILL., (+)-1-acetoxypinoresinol (11) and (+)-1-acetoxypinoresinol-4'-β-D-glucoside (1o), and by coumarin components of bark of Fraxinus japonica BLUME, scopoletin (6b), isofraxidin (7b) and fraxin (7e). The structure-inhibitory activity relationships were also discussed.
