Screening of Antiallergic Effect in Traditional Medicinal Drugs and Active Constituents of Aurantii Fructus Immaturus

Screening of antiallergic drugs used in traditional medicine was made on the basis of information given in literature. About one half of the selected samples showed inhibitory effect on rat passive cutaneous anaphylaxis (PCA). One of the most effective drugs, Aurantii Fructus Immaturus (枳実), was subjected to further separation with step-by-step monitoring of the inhibitory action of histamine release from rat peritoneal mast cells. Samples sparingly soluble in buffers were made into solutions by the aid of polyvinypyrrolidone (PVP). The fractions showing the strongest effect were found to contain three polymethoxyfiavones, nobiletin, 3, 5, 6, 7, 8, 3' 4'-heptamethoxyflavone and tangeretin, which were isolated as antiallergic constituents of this crude drug. The effect of tangeretin was less than that of the other flavonoids. Nobiletin inhibited PCA when orally administrated. Inhibition of nobiletin on the histamine release induced by concanavalin A (Con A) seemed to be due to its effect on Con A-independent stage.
