Identification of chromosomes in two Chinese spruce species by multicolor fluorescence in situ hybridization

Chromosomes of Picea koraiensis Nakai and P. crassifolia Komarov were analyzed by multicolor fluorescence in situ hybridization (FISH) using three repetitive probes, 45S rDNA, 5S rDNA and centromeric Sau3A family DNA. 45S rDNA signals appeared at interstitial regions of four or five pairs of long metacentric chromosomes. 5S rDNA signals were located at interstitial region adjacent to 45S rDNA locus of one chromosome pair which had very weak signal at the end of the long arm. A Sau3A repetitive DNA was located at the centromeric region of three pairs of long metacentric chromosomes in both species. The pattern of multicolor FISH with three probes could easily identify individual chromosome in both species and was very similar between the two species with differences in 45S rRNA loci.
