Comparison of 5S ribosomal DNA sites on somatic chromosomes of eight species in Cycas(Cycadaceae)

Somatic chromosomes of Cycas revoluta, C. taitungensis, C. wadei, C. siamensis, C. circinalis, C. media, C. micronesica and C. thouarsii were investigated by the florescent in situ hybridization method (FISH) using 5S ribosomal DNA (5S rDNA) probes. The chromosome number of the eight species was commonly 2n=22. In FISH, C. revoluta, C. taitungensis and C. wadei showed a 5S rDNA site on the interstitial region near the terminal in two t chromosomes ; the other five species of C. siamensis, C. circinalis, C. media, C. micronesica and C. thouarsii showed a 5S rDNA site on the interstitial region near the terminal in two t chromosomes and on the interstitial region near the centromere in the other two t chromosomes. The cytological data suggests that C. revoluta, C. taitungensis and C. wadei are cytotaxonomically the most similar to each other among the eight Cycas species investigated. The cytological relationships of the eight Cycas species in the present study corroborate morphological relationships postulated by Hill (1995, 2003).
