Marking and identification of nucleolar chromosomes in Allium grayi tetraploids by Ag-NOR staining and fluorescence in situ hybridization

Allium grayi Regel collected in Harbin, Heilongjiang Province, China, in Tonghae. Kangwondo, Republic of Korea and in Ohyamazaki Town. Kyoto Pref., Japan, were karyologically investigated. All the plants in Harbin and Tonghae showed the common chromosome number of 2n=4x=32, whereas the plants in Ohyamazaki showed the chromosome number of 2n=4x=32 and of 2n=5x=40. Karyotypes of the Tonghae and Ohyamazaki tetraploid plants and the majority of Harbin pants of the species were cornmonly, 2n=32=26m+5sm+1st, whereas those of a few plants in Harbin were 2n=32=26m+6sm. Ag-NOR staining and fluorescence in situ hybridization (FISH) detected that four among the six nucleolar chromosomes were median-centromeric and two were submedian-centromeric chromosomes.
