Discrimination and isolation of terminal chromosomal regions of Dendranthema occidentali-japonense in the chromosomes of F_1 hybrid between D. occidentali-japonense and D. boreale by using GISH

Artificial reciprocal crosses between Dendranthema occidentali-japonense (2n=54, hexaploid) and D. boreale (2n=18, diploid) were made and produced F_1 hybrid (2n=36, tetraploid). Applying DAPI on metaphase chromosomes of the hybrid showed the distribution of positive DAPI bands of Adenine-Thymine (A-T) rich heterochromatin regions located at the terminal regions of approximately 3/4 of the chromosomes of the complement, that exhibited prominent yellow-green GISH bands after in situ hybridization with the biotin-labelled total genomic DNA probe of D. occidentali-japonense. The genomes of the two species showed closely relationship by GISH since the chromosomes of the hybrid fluoresced yellow-green-color, that decreased the coloration with increasing the concentration of blocking DNA, excepting the terminal part of the chromosomes exhibited prominent signals of GISH after hybridization with the probe of D. occidentali-japonense. Inter- and/or intra-chromosomal rearrangement as inversion and/or inversion-translocation-recombination has been observed after GISH as interstitial positive GISH bands in some chromosomes, fluoresced yellow-green-color after hybridization with the probe of D. occidentali-japonense. Applying the reciprocal experiment using the biotinlabelled total genomic DNA of D. boreale as a probe, all of the chromosomes in the hybrid fluoresced yellow after hybridization with the probe, excepting the terminal region of the chromosomes showed the negative result of the DAPI-GISH bands and stained red
