Lysis of Rabbit γ-Globulin-Coupled Liposomes by the Complement System

The lysis of rabbit γ-glibulin (RGG)-coupled liposomes by the complement system was measured with and without the addition of anti-RGG antibody. The complement activity obtained from lysis with the anti-RGG antibody was well correlated with that of the CH50 hemolytic complement test (r=0.92). The degree of lysis without anti-RGG antibody was strongly affected by the addition of factor I and the pretreatment of serum with zymosan. Althouth the addition of C4 to C4-deficient serum remarkably increased the lysis with anti-RGG antibody, it had little affect on that without anti-RGG antibody. From these results, the lysis without anti-RGG seemed to be caused by the alternative pathway, and thus RGG-coupled liposomes can be used for measuring the activities of both the total and alternative pathway of complement.
