Effective Purification of Bioproducts by Fast Flow Affinity Chromatography

The performance of affinity adsorbents, such as HiPAC, Toyopearl, and Sepharose 4B coupled with protein A and anti-BSA, was compared. The HiPAC column, which is packed with rigid porous silica particles of 30 μm diameter, showed the highest value of D_<pore>/r^2 because of the small particle diameter and rigidity of the adsorbent particles. Further, it was operable at higher flow rates than the others. Thus, experimental data and calculation on the purification process show that fast and efficient purification is possible by use of the HiPAC column.
