Selective Production of 5-Keto-D-Gluconate by Gluconobacter Strains

Gluconobacter suboxydans IFO 12528 was selected as a strain performing 5-keto-D-gluconate (5KGA) production with the highest yield among Gluconobacter strains which produce both 2-keto-D-gluconate (2KGA) and 5KGA. This strain accumulated 5KGA with a yield of more than 90% of the initial carbon source during growth on a medium containing both glucose and gluconate, and also in the resting cell system. The productivity of 5KGA was higher with cells from the stationary phase than that of the midexponential phase. The optimum pH and temperature for 5KGA production by resting cells were found to be 2.5-3.0 and 20℃, respectively. For the selective production of 5KGA, the resting cell system was better than cultivation, since the production of 2KGA could be repressed in the resting cell system.
