A Novel Enzyme System : The n-Alkane-2-One-Forming Enzyme System in Penicillium decumbens IFO-7091

The enzyme system catalyzing n-nonane-2-one(2-one)formation from capric acid in Penicillum decumbens IFO 7091 was investigated. The enzyme system, which was constitutively formed, could be recovered in cell-homogenate. By fractionation of the homogenate, the system was separated into catalytic and cooperative units. The former unit was specifically located in the wall-membrane fraction(P_1)and the latter in the cytosol(S_2)fraction. Individually, the fractions did not have significant 2-one-forming activity; however, reconstruction of the enzyme activity was achieved by addition of S_2 to P_1 with full restoration. EDTA was a strong inhibitor of the enzyme activity, and it was proved that S_2 could be replaced with Ca^<2+>.
