Improvement of Cytochrome c Production by Glycine-and Glycine Analog-Resistant Mutants of Methylomonas sp.

• The cytochrome c production of glycine-resistant mutants of Methylomonas sp. YK 56 increased gradually with the increase of resistance to glycine. The strain AR 67,which was derived finally as a glycine analog-resistant mutant, had 2.3 times the cytochrome c production of strain YK 56. The methanol concentration and aeration volume affected the cytochrome c production. Under the optimal conditions, 103.5 mg of cytochrome c per l of culture broth was obtained.The activity of δ-aminolevulinate (ALA) synthase increased about 20-fold over that of strain YK 56,and the ALA content and activity of ALA synthase of strain AR 67 reached the maximum value at an early step of the cultivation. Three types of soluble cytochromes c of strain AR 67 were obtained, the same as strain YK 56. The amount of cytochrome c-III decreased according to the increase of the cultivation time.
