Efficiengt Introduction of Vector Plasmids into Acetic Acid Bacteria

An efficient and convenient method to introduce foreign plasmid DNA into acetic acid bacteria is by conjugal transfer. Broad host range plasmid vectors, RP4 and pRK290,were introduced into Accetobacter aceti K1006 from Escherichia coli as donor, and maintained stably. The conjugal transfer frequencies per recipient were 50-70% with RP4,and 2-20% with pRK290.Plasmid DNA transformation with pRK290 was also examined using calcium chloride treatment. Although we tried various modifiocations, the frequency was below 10^2 transformants per μg DNA.Our confugal transfer method was used on five strains of various species of Acetobacter, and frequencies of 0.1-1.5% were obtained in three strains. It is suggested that vector plasmids can be efficiently introduced into many acetic acid bacteria by this method, which would be useful for gene manipulation and lead to the development of genetic analysis in acetic acid bacteria.
