A method for obtaining pure cultures of Spirulina platensis was established, which consisted of (1) mechanical fragmentation of washed multicellular filaments in a homogenizer, (2) ultraviolet irradiation and (3) drop-by-drop inoculation to a number of tubes with adequately diluted inoculum. Cultural characteristics of photoautotrophic pure cultures were shown to be very similar to those observed previously in cultures contaminated with bacteria. The highest specific growth rate observed at 25-30 klux was 1.8 day^<-1>. Growth rates in the logarithmic as well as in the linear phase were generally a little higher in pure cultures than those in contaminated ones. S. platensis, like other blue-green algae, was shown to be an obligate photoautotroph. Mixotrophic cultures could be performed effectively to obtain remarkably higher cell yields and growth rates than simple photoautotrophic cultures. An interesting phenomenon of "mixotrophic lysis" was observed in cultures with added glucose when inoculum sizes were small.
