Requirement of Oxygen for Dark Hydrogen Evolution by a Green Alga, Chlamydomonas reinhardtii

External factors affecting dark hydrogen evolution of Chlamydomonas reinhardtii were investigated in order to utilize green algae in a two-stage biophotolysis system in which O_2 and H_2 evolutions are separated temporally. Hydrogen evolution was observed when a culture of C. reinhardtii was shaken under dark microaerobic conditions. The dark hydrogen evolution was inhibited by the addition of dithionite as an oxygen scavenger or uncoupler 2,4-dinitrophenol. Hydrogen-evolving activity acquired by the cells increased with the increase of O_2 tension up to about 0.23%. The addition of small amount of O_2 was also required for the acquisition of hydrogenase activity.Therefore, it was concluded that C. reinhardtii can not acquire the capability to evolve H_2 under strict anaerobic conditions, and addition of a small amount of O_2 is essential to the adaptation.It was demonstrated that C. reinhardtii evolved H_2 repeatedly in an alternating light/dark cycle, and a temporal two-stage biophotolysis proved feasible by use of this green alga.
