Purification and Characterization of α-Galactosidase from the Yeast Trichosporon cutaneum JCM 2947

α-Galactosidase was purified to homogeneity from the culture fluid of the yeast Trichosporon cutaneum JCM 2947 (=T. beigelii CBS 5790) by column chromatography on DEAE-Toyopearl 650M, Butyl-Toyopearl 650M, hydroxylapatite and Sephacryl S-200. The molecular weight of the purified enzyme was estimated to be 85,700 by SDS-polyacrylamide gel electrophoresis and 321,000 by gel filtration. Optimum pH and temperature were 5.0 to 6.0 and 45℃, respectively. The K_m (mM) for p-, o-, m-nitrophenyl α-D-galactopyranoside, melibiose, raffinose, and stachyose were 0.14, 0.25, 0.49, 6.0, 48, and 68, respectively, and V_<max>. (μmol/min/mg protein) for those substrates were 66.2, 22.6, 12.6, 12.6, 28.9, and 18.0, respectively.
