Plasmid pUO2 Determining Haloacetate Dehalogenase and Mercury Resistance in Pseudomonas sp.

Pseudomonas sp. strain C, a strain capable of assimilating haloacetate, was found to harbor a plasmid, pUO2,which determines the production of two haloacetate dehalogenases, H-1 and H-2,as well as mercury resistance. A spontaneous H-2 deficient mutant of strain C was found to harbor a deletion plasmid, pUO21. pUO2 and pUO21 were respectively very similar to pUO1 and pUO11,which had been found in Moraxella sp. strain B. Cleavage analysis of plasmid DNAs with restriction enzymes revealed that pUO2 molecule was similar to, but about 0.7 Mdal shorter than pUO1 (43.7 Mdal), and that pUO21 and pUO11 molecules lacked identical DNA fragments (about 3.6 Mdal in sum). Moraxella B produced H-1 inducibly and H-2 constitutively, whereas Pseudomonas C produced both enzymes consitutively and in a higher ratio of H-2 to H-1. Conjugational transfer of pUO2 from Pseudomonas C to Moraxella B (plasmid-free) revealed that the regulation of dehalognase production was host-dependent.
