Purification and Some Properties of an Endo-1,4-β-D-Xylanase from Streptomyces sp.

A strain of Streptomyces sp., tentatively named Streptomyces sp. KT-23,was isolated from rotten rice-straw and found to produce an extracellular xylanse. The xylanase was purified to homogeneity by chromatographies on DEAE-cellulose and Bio-Gel P-100. The purified enzyme had an apparent molecular weight of 43,000 and its isoelectric point was at pH 6.9. The pH and temperature optima for the activity were 5.5 and 55℃. The enzyme acted on β(1-4)-D-xylan to produce mainly xylobiose and a small amount of xylose as final products. Its pattern of action on β(1-4)-D-xylan indicates that it is an endo-1,4-β-D-xylanase (EC 3.2.1.8).
