An α-Glucan from Suspension-Cultured Rice Cells

An α-glucan was isolated from 11-day-old suspension-cultured rice cells by extraction with hot Na-phosphate buffer (pH 6.8). The α-glucan had [α]_D= +234° (C=0.14, in water) and its average molecular weight was estimated to be about 1.4 × 10^4, based on elution characteristics on a calibrated Sepharose CL-6B column. Upon partial acid hydrolysis, the α-glucan gave mainly malto-oligosaccharides. The maximum absorption of the iodine complex of the α-glucan in the presence of Na_2SO_4 was at 470 nm. The results of hydrolysis by α-, β- and iso-amylases and methylation analysis indicated that the isolated α-glucan is a highly branched polysaccharide with an average chain length of 9. The exterior and interior chain lengths of the α-glucan were calculated to be 5 and 3, respectively.
