Isolation of Xyloglucans from etiolated Glycine max and Vigna sesquipedalis hypocotyls

Xyloglucans isolated from cell walls of etiolated Glycine max and Vigna sesquipedalis hypocotyls were subjected to fragmentation analysis with cellulase for structural comparison with those derived from Phaseolus aureus hypocotyls. The xyloglucans from G. max and V. sesquipedalis had glucose, xylose, galactose and fucose in the approximate molar ratio of 10 : 6 : 4 : 1 and 10 : 7 : 3 : 1 , respectively. However, the results of cellulase fragmentation analysis of xyloglucans from the three species suggested that the basic structure of the xyloglucans in the cell walls of these bean-hypocotyls is almost the same; the structure is based on two repeating oligosaccharide units, one of which consists of glucose and xylose and the other of glucose, xylose, galactose and fucose.
