Molecular Cloning and Expression of β-Isopropylmalate Dehydrogenase Gene from Alkalophilic Bacillus in Escherichia coli

The gene for β-isopropylmalate dehydrogenase of alkalophilic Bacillus no. 38-2 has been cloned into the HindIII site of pBR322 and expressed in E. coli. The plasmid pHK201,containing a 5.7-kb HindIII fragment, was isolated from the leucine^+ transformants. Genomic hybridization showed that the 5.7-kb HindIII fragment was derived from the chromosomal DNA of alkalophilic Bacillus no. 38-2. Restriction analysis showed that β-isopropylmalate dehydrogenase gene was located in the 1.1-kb HindIII-HincII fragment.
