Bone Morphogenetic Protein (BMP) Stimulates Cytological and Functional Differentiation of Preodontoblasts in vitro

Bone morphogenetic protein (BMP) was partially purified from bovine bone and characterized biochemically. This BMP fraction contained BMP-2 and contaminants, such as histones H3 and H2B, and was free of basic fibroblast growth factor and transforming growth factor-β1 and-β2. The BMP fractlon, either alone or combined with heparin, promoted functional differentiation of odontoblast-like cells and stlmulated matrix secretion in cultured first lower molar dental papillae isolated from day-17 mouse embryos. The induction of polarization and functioral differentiation of odontoblast-like cells were dependent on the dose of the BMP fraction. Interestingly, the initiatlon of odontoblast-like cen differentiation with maintenance of physiological gradients could be stlmulated ir the presence of BMP-2 and other components even without heparin. These results demonstrate that BMP-2 the stimulates the cytological and functional differentiation of preodontoblasts, and that other components might play important roles as a substrate (similar to heparin), and/or as a synergistic cofactor of BMP-2.
