The Effect of Enzymatic Treatment on Immunohistochemical Staining of Collagens in Mandibular Condyle of Rats

This study was performed to evaluate the effect of enzymatic treatment on immunohistochemical localization of types I and II collagen in mandibular condyle of rats. The 4% paraformaldehyde and 0.1% glutaraldehyde-fixed, paraffin-embedded tissue sections were stained by anti-type I and II collagen antibodies using the PAP method. In order to compare the immunostaining conditions under various enzymatic treatments, the sections were digested with hyaluronidase, pepsin, trypsin, and actinase. The results demonstrate a variety of immunohistochemical reactions for each collagen according to the applied enzymes. The specificity and intensity of immunostaining for typesI and II collagen in the mandibular condyle were more favorable in pepsin treated sections than in other section.
