Principles of Electron Spectroscopic Imaging(ESI)in Hard Tissue Biology

The main signal due to inelastically scattered electrons from biological soft and hard tissues can be analyzed by electron spectroscopic imaging (ESI). lnelastically scattered electrons are produced when an incident electron beam penetrates a thin biological specimen composed of light elements. The inelastically scattered electrons include plaslnon-loss electrons, core-loss electrons, and background electrous. ESI is able not only to detect the weak signal due to phosphorus in cyioplasmic membranes and ribosomes but also to map the elements. Fresh biological tissue can produce 50 times more inelastically acattered electrons than standard specimens processed by double fixation and double staining. Biological specimens that can be analyzed by ESI include plastic-embedded standard sections after double staining, cryo-sections fixed in glutaraldehyde, fresh cryo-sections of human hair, negatively stained erythrocyte membranes treated with streptolysin O, ferritin particles on a supporting film of osmium-oxide polymer, and biologically synthesized hydroxyapatite crystals. These preparations can be observed by use of various imaging procedures, such as zero-loss imaging, plasmon-loss imaging, core-loss imaging, 250-eV loss imaging, or filtered electron beam diffraction. Elemental mapping of carbon and iron in ferritin molecules is also possible. An imaging plate is one hundred times more sensitive than common films used for electron maicroscopy. Therefore, a natural image can be recorded before the specimen is damaged by the incident beam of electrons. Imaging filters with fast Fourier transfer (FFT) are powerful tools and allow extraction of detailed profiles from noisy images.
