The Effect of BMP on the Monocyte-Macrophage-Mesenchymal Cell Interaction in Ectopic Bone Formation

• Partially purified bovine bone morphogenetic protein (BMP) absorbed in fibrous glass membrane (FGM) was implanted to the dorsal subcutaneous tissue of rats to investigate the effect of BMP on the monocytes/macrophages in ectopic bone formation. FGM alone was implanted as a control. The implants were harvested at 1, 2, and 3 weeks after implantation, then prepared for routine light microscopic and electron microscopic observations. At 1 week with some formations of multinucleated giant cells, many macrophages showing a positive immunoreaction for ED1 Ab, as well as mesenchymal cells, migrated into the BMP-FGM complex. The recruited intact macrophages in the BMP-FGM complex revealed more abundant cytoplasmic organelles and aclose intercytcplasmic relationship with the intermingling proliferated mesenchymal cells. However macrophages in the control showed less cellular activity due to the toxicity of FGM. These findings support the suggestion that BMP may serve as a potent chemoattractant to recruit monocytes as well as mesenchymal stem cells, and their close relationship may act synergistically to promote the bone inductive cascade.
