Gene Expression of Chondro-Osseous Cells in Heterotopic Bone formation Induced by rhBMP-2

The in situ hybridization method was applied to observe the heterofopic cartilage-like tissue induced by recombinant human bone morphogenetic protein-2(rhBMP-2). The localization of collagen II protein, osteopontin (OPN) and osteocalcin (OCN) mRNAs was evaluated in rhBMP-2 induced tissue in comparision with the cartilage tissue in normal endochondral ossification mode of the rat fetus. In the heterotopic cartilage-like tissue, OPN and OCN mRNA were detected in chondrocyte-like cells; collagen II was noted in the cartilago-like matrix. In the cartilage tissue in the endochondral ossification of the fetal rat tibia, OCN mRNA was absent in chondrocytes. These results suggest that the chondrocyte-like cells in the BMP induced cartilage-like tissue have the function of osteoblasts. The chondrocyte-like cells in the inductive cartilage-like tissue may be designated as chondro-osseous cells instead of chondrocytes. The heterotopic chondro-osseous tissue formation induced by rhBMP-2 differs from normal endochondral ossification in cell differentiation and function.
