Aging and Ectopic Bone Formation Induced by Partially Purified Bone Morphogenetic Protein : Blood Vessel Ingrowth and Localization of Type I Collagen and Osteocalcin Assessed by Immunohistochemistry

A mixture of heparin-Sepharose-purified bovine bone morphogenetic protein (BMP) and type I atellocollagen as the carrier was implanted in the subcutaneous tissues of 4-week, 10-month and 18-month old rats. The explants were evaluated usilrg H&E staining, morphometric analysis, as well as immunohistochemical staining of type IV collagen for blood vessel distribution, type I collagen and ostcocalcin for bone matrix proteins. Ectopic bone formation was reduced and the rate of blood vessel ingrowth was delayed and hindered in aged rats. The difference in blood vessel distribution might be one reason for the reductlon in ectopic bone formation in aged rats. The localization of type I collagen and osteocalcin was consistent with the bone formation observed histologically, L e., more extensive areas positive to type I collagen and osteocalcin in 4-week old rats than in 10-month and 18-month old rats.
