Effects of Bovine Amelogenin and Enamelin on in vitro Mineralization in Gel

Amelogenins and enamelins are the major extracellular-matrix proteins in developing mammalian enamel. The effects ofthese proteins on mineralization were examined using an in vitro mineralization system in gel. Two proteins, 28kDa amelogenin and 70kDa enamelin, were purified from bovine enamel. Gels containing these proteins were used as diffusion media for calcium and phosphate ions. Calcium phosphate was precipitated in the gels, and was identified as octacalcium phosphate by X-ray and electron diffraction. The precipitation band was sharp in the gel containing the enamelin, although it was broad in the gel containing the amelogenin. Electron microscopic observation showed that crystals precipitating in the former gels were 2-5 times larger than crystals in the later gels. Hydroxyapatite chromatography of these two protcins indicated that the enemelin had a higher affinity to the mineral than the amelogenin. Amelogenin may be responsible for the diffuse nucleation of the crystal, and enamelin may facilitate the localized nucleation and regulate crystal growth.
