In morphological studies of the basement membrane, the use of a marker provides a valuable method. Recently, the authors extracted type IV collagen from a bovine lens to be used as an antigen for the preparation of a marker antibody to type IV collagen, a component of the basement membrane. In the present study the bovine lens capsule was pepsinated and subjected to salt-differential precipitation for isolation and purification of type IV collagen. Type IV collagen thus obtained was identified with sodium dodecyl sulfate-polyacrylamide gel eletrophoresis (SDS-PAGE) and amino acid analysis. By using this as an antigen, an antibody to type IV collagen was prepared in rabbits, and subsequently purified with affinity chromatography. When renal tissues of rats were immunohistologically examined with this antibody, positive reactions were found in the tubular basement membrane, the interstitial capillary basement membrane, the mesangium and Bowman's capsule. In the basement membrane of the glomerular capillary vessels, however, a positive reaction was found in only a part of the membrane and was very weak. These results indicate that type IV collagen in the basement membrane has different immunological properties related to different functions of the membrane.
