The "Microdialysis system" was investigated to determine the amount of tissue damage caused by insertion of the dialysis probe. A microdialysis probe was inserted into the frontoparietal cortex of the Sprague-Dawley rats anesthetised with pentobarbital. The monoamine concentration in the perfusate was measured by HPLC. Horseradish peroxidase (HRP) was administered intravenously 15 minutes before the rats were sacrificed. The findings were : (1) In general, the monoamine concentration was highest in the initial perfusate, and decreased thereafter. (2) HRP was extravasated at the probe insertion site. (3) Edematous and necrotic changes were observed around the probe wound in a region from 400 to 800 μm in diameter. In addition, 5/12 (42%) of the rats showed secondary lesions branching from the probe wound. It was hence concluded that the Microdialysis system caused considerable local tissue damage. Therefore, the data obtained with this apparatus must be discussed by taking into account the dynamic equiliblium between extravasated edema fluid and the cerebral interstitial fluid in acute experiments.
